what is PNC-27? function?

PNC-27 is a synthetic peptide that has been researched for its potential anti-cancer properties.


PNC-27 is a designer peptide composed of:

  • A sequence from the p53 protein (a well-known tumor suppressor),

  • Fused with a membrane-translocating leader sequence (helps the peptide enter cells).

It was originally developed in research led by Dr. Matthew Pincus and others at SUNY Downstate Medical Center.


Function and Mechanism

Primary Proposed Function:

PNC-27 is designed to target and kill cancer cells by:

  1. Binding to HDM-2 (also called MDM2), a protein that inhibits p53.

  2. Inserting into the plasma membrane of cancer cells, forming pores.

  3. Disrupting the membrane integrity, leading to necrosis (cell death).

Importantly, it has been reported to:

  • Kill cancer cells (in vitro and in some animal models),

  • Spare normal, healthy cells.


Status and Controversy

  • PNC-27 has not been approved by the FDA or any major regulatory body.

  • It is not an established cancer therapy in mainstream medicine.

  • Some clinics, especially outside the U.S., have offered it as an experimental or alternative cancer treatment, but scientific validation is lacking.

  • There are no large-scale clinical trials proving its safety or efficacy in humans.
